Glycoforms In Health & Disease - The Science

  • The glycans on glycoproteins contribute to protein function through properties such as stability and interactions.

  • All current tumor markers (e.g. CA125, PSA) are glycoproteins but current assays do not measure the glycans on the proteins. Studies show improved accuracy when glycoforms are measured.

  • Altered glycans and glycoforms are observed during the development of diseases such cancers, inflammatory and autoimmune conditions.

Glycoform = a variant of glycoprotein defined by the particular sugars.

Glycan = scientific term for sugar.

Design Features

01 

Streamlined glycoform enrichment with in-built quality control

  • Spiked internal control protein added to each serum sample for quality assurance during LeMBA.

  •  Liquid handler pulldown and on-bead tryptic digest for throughput and consistency.

Single platform from research to clinical 

LeMBA is coupled to appropriate MS techniques: data-dependent acquisition (DDA) or data-independent acquisition (DIA) for discovery, multiple reaction monitoring (MRM) for validation and clinical assay.

Rapid deployment to clinical MS assay 

  • No new reagent to be developed.

  • Reduces the time, cost and risk of product development.

04 

Diagnostic panel with algorithm increases accuracy 

  • Algorithm will be developed to interpret probability of ovarian cancer.

Proof of concept completed: esophageal adenocarcinoma biomarkers licensed to Proteomics International in June 2022.
